Indications/Uses
Zypezil 5/Zypezil 10 Symptomatic treatment of mild to moderately severe Alzheimer's dementia. Zypezil ODT 10 Treatment of dementia of the Alzheimer's type. Mild, moderate & severe Alzheimer's disease.
Dosage/Direction for Use
Zypezil 5/Zypezil 10 Adult/elderly Initially 5 mg/day & should be maintained for 1 mth. May be increased to 10 mg/day after 1 mth of clinical assessment of treatment. Max: 10 mg daily. Zypezil ODT 10 Mild to moderate Alzheimer's disease 5-10 mg once per day. Severe Alzheimer's disease 10 mg. Recommended starting dose: 5 mg once daily.
Administration
May be taken with or without food: FC tab: Take at bedtime. ODT 10: Allow to dissolve on the tongue & follow w/ water.
Contraindications
Hypersensitivity to donepezil HCl or piperidine derivatives.
Special Precautions
Exaggerate succinylcholine-type muscle relaxation during anaesth. Pregnancy & lactation. Not recommended for childn. Zypezil 5/Zypezil 10 Patients w/ severe Alzheimer's dementia, other types of dementia or other types of memory impairment (eg, age-related cognitive decline). Exaggerate succinylcholine-type muscle relaxation during anaesth. Sick sinus syndrome or other supraventricular cardiac conduction conditions eg, SA or AV block. Patients at increased risk of developing ulcers eg, those w/ history of ulcer disease or those receiving concurrent NSAIDs should be monitored for symptoms. May cause bladder outflow obstruction. Exacerbate or induce extrapyramidal symptoms. Patients w/ history of asthma or obstructive pulmonary disease. Avoid concomitant use w/ other inhibitors of acetylcholinesterase, agonists or antagonists of the cholinergic system. Galactose intolerance, Lapp lactase deficiency or glucose-galactose malabsorption. Minor or moderate influence on the ability to drive & use machines. Severe hepatic impairment. Zypezil ODT 10 Bradycardia or heart block in patients w/ or w/o underlying cardiac conduction abnormalities. Nausea, vomiting, diarrhea. PUD & GI bleeding. Seizures. Patients w/ history of asthma or obstructive pulmonary disease.
Adverse Reactions
Diarrhoea, muscle cramps, fatigue, nausea, vomiting & insomnia.
Drug Interactions
Inhibited metabolism w/ ketoconazole, quinidine, itraconazole, erythromycin & fluoxetine. Increased mean conc w/ ketoconazole. May reduce levels w/ rifampicin, phenytoin, carbamazepine & alcohol. Potential to interfere w/ medications having anticholinergic activity. Potential for synergistic activity w/ succinylcholine, other neuromuscular blocking agents or cholinergic agents or β-blocking agents.
